We are working to refine an integrated MS-based workflow for intact, native complexes, i.e. "complex-down" characterization, with state-of-the-art instrumentation and software that can answer structural biology questions. An integrated workflow for native MS defines the m/z of the intact complex, m/z of subcomplexes, stoichiometry, heterogeneity, connectivity of partners, topology, conformational diversity, collision cross sections, and relative subunit binding strengths of intact, native protein complexes with high sensitivity and throughput. The workflow integrates three Technology Optimization Projects (TOPs).

nMS ion clean-up, conformational control, and thermodynamic measurements

Higher resolution m/z selection, FT-ion mobility, surface collisions, and computations

Ion Activation, unfolding, and dissocation strategies for native proteomics and protein quaternary structure discovery
